Overview:
Timely and accurate pregnancy detection in ruminants—such as cattle, goats, and sheep—is vital for effective herd reproductive management. Early identification of non-pregnant animals allows for prompt rebreeding, reducing calving intervals and enhancing overall farm productivity.
This rapid test employs a double-antibody sandwich method combined with colloidal gold chromatography to detect pregnancy-associated glycoproteins (PAGs), which are specific markers produced by the placenta during pregnancy.
- In dairy cows as early as 28 days post-breeding and for cows over 60 days postpartum
- In goats starting from 28 days post-breeding
- In sheep from 35 days post-breeding
Detection Principle:
Upon adding the sample to the test card, any PAGs present bind to gold-labeled antibodies, forming a complex. This complex migrates along the test strip and is captured by antibodies immobilized on the test line (T line), resulting in a visible red band. The intensity of the band correlates with the concentration of PAGs. A control line (C line) should always appear to confirm the test's validity.
Test Procedure:

Preparation:
- Ensure all test components and samples are at room temperature (15–35°C) before starting.

Sample Collection:
- Whole Blood: Collect blood in EDTA tubes and mix gently by inverting the tube 10 times. Store at 2–8°C for up to 48 hours. Do not freeze.
- Serum/Plasma: Centrifuge collected blood at 2000–3000 rpm for 5–15 minutes or allow it to stand until separation occurs. Store at 2–8°C for up to 7 days.

Testing:
- Open the foil pouch and place the test card on a flat surface.
- Apply 2 drops (~50 µL) of the sample to the sample well.
- Add 2 drops (~50 µL) of sample diluent to the same well.
- Start a timer for 20 minutes.

Interpret the results:
- Positive: Both T and C lines appear.
- Negative: Only the C line appears.
- Invalid: No lines appear or only the T line appears.
Note: Even a faint T line indicates a positive result.
